Allegedly peptides are so finicky that you may only store them right at the back of the fridge.

Therefore here is the research on SEMAX Stability – a great substance that releases BDNF and regenerates the brain.

As a young doctor he worked in a nursing home / care home and there met an old Russian woman. She was relatively pre-demented and 80 years old and in a rather poor condition.

BPC-157 repairs my hip joint

Unfortunately, as a physician one is not allowed to use “peptides” on patients because they have no approval,

but in a heroic self-experiment I can report that my hip, which was severely impaired by a labral tear as well as subchondral cysts of the joint cartilage and for 3 years allowed me no more than a limping walking distance of 200 m, was – honestly speaking – completely healed with BPC-157 within a few weeks.

No wonder that BPC-157 is banned as a DOPING AGENT!

Peptides

I am especially interested in SEMAX because of its neuroregenerative activity
Therefore the following research on stability.

 

they told us that peptides do not stay stable even in lyophilized state

“… because if you open the fridge door and they warm up even slightly, they fall apart even in the lyophilized state …. was explained to us during the peptide training“.

Now this contradicts everything we used to do in the research lab, where we used diluted and sterile-filtered albumin solutions for months in the fridge, or lactoferrin or other proteins that we prepared ourselves.

 

This is how the Russians do it with SEMAX

The Russian pharmaceutical form is remarkably simple. The currently registered SEMAX 0.1 % nasal product from Peptogen contains per liter:

  • SEMAX 1 g
  • Methylparahydroxybenzoate, Nipagin, 1 g
  • purified water ad 1 liter

So converted:

  • SEMAX 1 mg/ml
  • Methylparaben 1 mg/ml = 0.1 %
  • purified water

There is no classical phosphate, citrate or acetate buffer indicated, also no benzyl alcohol and no NaCl. (Vidal Russia)

 

The decisive point

The Russians therefore apparently do not stabilize SEMAX by means of a complicated buffer, but essentially by:

SEMAX + 0.1 % methylparaben + water + refrigerated storage.

The official Russian medicinal product information states storage of the preparation at 2-8 degrees C. (Vidal Russia)

This also matches the manufacturer information: SEMAX contains a preservative and can therefore remain at room temperature for a few days; for longer storage, however, the manufacturer recommends the refrigerator. (Semax)

 

This practically refutes the claim …..

“SEMAX falls apart immediately as soon as it is dissolved”

practically refuted.

The Russian finished medicinal product is already an aqueous SEMAX solution, is sold as a multi-dose bottle and possesses pharmaceutical shelf life. It can therefore obviously be formulated sufficiently stably in the aqueous phase. (Vidal Russia)

Interestingly, a slightly acidic pH range of around pH 4.0-5.5 is described for aqueous SEMAX. This could be an essential part of the chemical stability, even if the current specialist information does not list a separate buffer. (medbiol.ru)

 

If one wanted to replicate the Russian formulation

For an intranasal pharmaceutical formulation the Russian principle would therefore be:

SEMAX 1 mg/ml + methylparaben 1 mg/ml + purified water, refrigerated at 2-8 degrees C.

This is considerably closer to the original than bacteriostatic water with 0.9 % benzyl alcohol. Benzyl alcohol is precisely not contained in the Russian SEMAX recipe. (Vidal Russia)

An important caveat: I would not automatically derive a recipe for sterile injection from this. The Russian formulation is explicitly intranasal, and the requirements for sterility, endotoxins, osmolality and excipients are different for parenteral applications.

 

Even more interesting would be the original development

I can go even deeper into the Russian SEMAX patents and pharmaceutical development work. There one might find why exactly 0.1 % Nipagin, which pH range and which measured residual SEMAX concentration after months were chosen. That would be the actual answer to the question of how SEMAX is chemically maximally stabilized.

 

Methylparaben in Semax – any effect other than bactericidal action?

Primarily methylparaben is a preservative. It is intended to prevent the growth of bacteria and especially yeasts/fungi in the aqueous multi-dose solution. A relevant direct chemical stabilization of the SEMAX peptide by methylparaben I would not assume.

 

What it achieves indirectly

Methylparaben can improve shelf life indirectly:

  • prevents microbial degradation of the peptide
  • prevents pH changes caused by microorganisms
  • enables the repeated use of an aqueous multi-dose bottle

But it does not specifically prevent the typical chemical degradation pathways of a peptide such as oxidation, hydrolysis or deamidation.

 

The interesting thing about the Russian recipe

This leaves the actually exciting question:

Why is SEMAX nevertheless sufficiently stable in the Russian aqueous solution?

The more probable explanation is that SEMAX itself in clean water at a suitable temperature is considerably more stable than the claim “immediately unstable after reconstitution” would suggest.

The Russian finished formulation is a fairly strong practical counter-argument for this: It consists essentially of SEMAX + water + antimicrobial preservative, without an obvious classical stabilization system.

I would therefore functionally classify methylparaben as follows:

Methylparaben = microbial preservation, not actual peptide stabilization.

For the chemical stability of SEMAX, temperature, pH, oxygen/light and possibly the oxidation of the N-terminal methionine are presumably considerably more interesting.
